Immatics Highlights Compelling Anti-Tumor Activity of Anzu-cel PRAME Cell Therapy in Metastatic Uveal Melanoma at the ESMO 2025 Presidential Symposium

A Cure In Sight’s Research Coordinator (Kevin) is attending the European Society for Medical Oncology meeting in Berlin. Dr. Sapna Patel, from the University of Colorado, presented in the closing Presidential Symposium the updated results from of a Phase 1 study evaluating a single dose of a novel investigational immune therapy, anzutresgene autoleucel (anzu-cel, formerly IMA-203). Anzu-cel is a PRAME-directed T-cell therapy made from the patient’s own immune cells. These cells are modified in the laboratory so they can better identify the cancer-specific target PRAME. Once infused back to the patient, they can find and attack the cancer more precisely. They may stay for months to years in your body killing cancer cells. The Phase I portion of the study has so far enrolled 74 heavily-pretreated patients with advanced and metastatic solid tumors, who were HLA-A*02:01 positive and PRAME positive.

In this report, Dr. Patel focused on a subset analysis of 16 patients with metastatic uveal melanoma. These patients had an average of two prior therapies, including 62% who had prior tebentafusp. The most frequent adverse effects, occurring in almost all patients, included low blood counts and cytokine release syndrome (CRS). CRS was primarily low-grade, and mostly resolved within 14 days. All patients (16/16) experienced tumor shrinkage and two-thirds of patients achieved a response to treatment. Responses to anzu-cel therapy appear to be durable to almost one year.

The Phase II portion of the study is beginning to enroll patients (study numbers: NCT03686124, EudraCT 2019-002370-31), and the therapy’s sponsor Immatics is currently adding additional US cancer centers with uveal melanoma clinical trials experience. ACIS will provide updates on these new sites as the information is made available.

These emerging clinical data, although initially reporting a limited number of patients with metastatic UM, may be something you may wish to discuss with your medical oncologist.
